1. StyD4I is a neoschizomer of ScrFI.
2. StyD4I (CCNGG) has 4 possible recognition sequences: two, CCAGG andCCTGG can be blocked by dcm methylation CCWGG (W = A or T). The othertwo recognition sites CCCGG and CCGGG are not blocked by dcmmethylation. Note dcm methylation can be removed or avoided by using theNEB dam-/dcm- Competent E. coli strain (NEB #C2925)
3. Blocked by overlapping dcm methylation and impaired by overlapping CpG methylation.

1. When is StyD4I blocked by overlapping dcm methylation?
2. Do degenerate recognition sites need to be palindromic?
