1. Demonstrates marked site preferences and cuts pBR322 DNA very slowly.
2. Inhibited by salt concentrations > 50mM.
3. NgoMIV, an isoschizomer of NaeI, exhibits less site preference.
4. For enzymes that cannot be heat-inactivated, we recommend using a column for cleanup (such as the Monarch® PCR & DNA Cleanup Kit), or running the reaction on an agarose gel and then extracting the DNA (we recommend Monarch Gel Extraction Kit), or performing a phenol/chloroform extraction.
5. Requires two or more sites for cleavage. Please review the enzyme list and recommendations in the following table as well as the background information article.
6. Blocked by CpG methylation.

1. I tested your restriction enzyme on the substrate DNA recommended by NEB, and it appears to be active, however it does not digest my DNA. What could be the reason?
