1. BseYI can remain bound to DNA after cutting and alter migration rate of DNA during electrophoresis. To disrupt binding, add SDS to a final concentration of 0.5% or purify DNA before electrophoresis.

1. Is BseYI activity sensitive to dam, dcm or mammalian CpG methylation?
2. How many base pairs should be added at the end of a PCR primer adjacent to the BseYI recognition site to guarantee that BseYI will cut properly?
