1. AciI has a non-palindromic recognition site. As a result, when DNA is cut with AciI and then ligated, only 50% of these ligated sites regenerate an AciI site.
2. This enzyme does not benefit from incubations over 1 hour. Visit "Restriction Endonucleases-Survival in a Reaction" for more information.
3. For enzymes that cannot be heat-inactivated, we recommend using a column for cleanup (such as the Monarch® PCR & DNA Cleanup Kit), or running the reaction on an agarose gel and then extracting the DNA (we recommend Monarch Gel Extraction Kit), or performing a phenol/chloroform extraction.
