1. SfcI concentrations of < 1 unit/µg DNA in a reaction are not recommended. 
2. SfcI is active for one hour in a reaction at 37°C, after one hour no further digestion takes place. Please refer to Restriction endonuclease survival in a reaction for more information regarding this topic.
3. The stability of SfcI is greatly enhanced by incubations at 25°C.Although the enzyme is only 25% as active at this temperature, we recommend an incubation temperature of 25°C for 4 - 16 hours when digesting supercoiled plasmids. At 25°C, 0.1 unit will digest 1µg of substrate DNA in 16 hours.
4. Not sensitive to CpG, dcm, or dam methylation.
5. star activity may result from a glycerol concentration of >5%.

1. Do degenerate recognition sites need to be palindromic?
2. I tested your restriction enzyme on the substrate DNA recommended by NEB, and it appears to be active, however it does not digest my DNA. What could be the reason?
