Small RNA Library Preparation
Small RNA (sRNA) has experienced a resurgence of interest over recent years, as more sRNA species have been identified, defined, and functionally resolved. Ranging from microRNAs (miRNAs) and small interfering RNAs (siRNAs) to small nucleolar RNAs (snoRNAs) and piwi-interacting RNAs (piRNAs), small RNAs have typically been considered to include any RNA under 200 nt in length.
Accurate library prep is essential for the identification and analysis of small RNAs, and NEBNext offers several kits to meet your analysis and workflow needs:
NEW! The NEBNext Low-bias Small RNA Library Prep Kit (NEB #E3420) is the next generation of small RNA library prep from New England Biolabs. This new kit features a fast, single-day workflow (~3.5 hours) from 0.5 ng – 1,000 ng of Total RNA (or 0.05 ng – 5 ng of Enriched Small RNA).
The NEBNext Low-bias Small RNA Library Prep Kit is supplied with the required adaptors, but indexed UDI primer pairs are available separately. NEB recommends using NEBNext LV Unique Dual Index Primer Pairs (Sets 1-5; NEB #E3400, E3402, E3390, E3392, E3404, E3406, E3408) for multiplexing up to 480 reactions.

The workflow of the first-generation NEBNext Small RNA Library Prep takes about one day, generating libraries from 1 ng – 1,000 ng of Total RNA. NEB recommends that existing users of one of the first-generation kits consider moving to the new, improved workflow of the NEBNext Low-bias Small RNA Library Prep Kit.
