The minimum amount of growth media from GLuc-expressing cells needed in a Western to detect GLuc activity will vary from cell line to cell line, and also will depend on the amount of GLuc expressed by the cells and the transfection efficiency. To start titering the amount of growth media that will be needed to detect GLuc in a Western by using the Anti-GLuc Antibody use a minimum of 0.1% of total growth media (from a high efficiency transfection). We suggest using 2-fold serial dilutions of the GLuc-containing growth medium as samples. Typically, one needs to use a minimum of 0.1% of total growth media from a high efficiency transfection.
